I'd like to call off the signing up for the trip.
I'd like to cancel my trip.
I'd like to call off the trip with you.

Hi,
I signed up for a trip with a local church this coming Sunday, but something comes up and I can't attend the trip. So I call the person in charge, but I'm not sure which one of the above should I use. Thanks for your enlightment in advance.

I'd like to call off the signing up for the trip.

Means that the signing itself is an ongoing process, and now you're canceling it.

I'd like to cancel my trip.

This one I like best of the three.

I'd like to call off the trip with you.

A bit complicated and formal to my ears.

If it were me calling, I'd simply say something along the lines of what you wrote in your description:

I'm sorry, but I can't attend the trip ...
