a. "Sally who is 25 years old is going to get her masters degree this year ".
b. "Sally, who is 25 years old, is going to to get her masters degree this year "

Are the commas necessary?
Does the first sentence mean that there is more than one person named Sally and we are trying to say which Sally is going to get her masters degree?

The commas are necessary. The first sentence would mean what you suggest, but I do not recommend it.
