Clark: How'd you get here?

Lois: Feminine charm. Yes, I do have some.

Clark: Great job of protecting your short supply of it.

Meaning you're doing well with the little you have left?

Lois: Why don't you give your stand-up a rest (Stop fooling around?)

and do exactly what I tell you?

1.  Really a quite nasty way of saying, "we would never know you had any (charm)".

2.  "Stand-up" refers to stand-up comedy, a common stage genre.  Here, 'stop trying to be funny'.
