Hi,

Should I say "I'll be back from Regina this evening", or "I'll be back in Regina this evening"?

You should say whichever is true!

If you were in Regina earlier and are now returning to the place you started (not Regina), then you will be back from Regina.
If you were somewhere other than Regina and are now returning to Regina, then you will be back in Regina.

The place after "from" tells where you went.  The place after "in" tells where you are or will be.
