Which one is correct?-- Both are OK.  No comma is necessary with short, appositive names.

(a) Where is my sister Jennifer?
(b) Where is my sister, Jennifer? (I choose this)

If the quotation is interrupted and then continues in your sentence, do not capitalize the second part of the quotation. These are OK:

(ii) "Please, " pleaded John, "tell me where my brother is. "
(iv)   "Please, " pleaded John, "tell me where my brother is. "

'The question is,' said Humpty Dumpty, 'which is to be master-- that's all.'

How about this?

"Please", pleaded John, "tell me where my brother is. "
