1) Do it right now.

2) Do it right away.
3) Do it at once.
Are there differences in meaning? Does (3) sound any harsher, or is it requiring a more immediate action by the listener?

right now is the harshest.  No excuses! Do it right now!
right away is closer to at the earliest possible opportunityWe're late.  We have to leave right away.
at once is simply (in the U.S.) a formal way of saying right away.
