If you are citing a direct quote from a book, and inside that, there is another quotation, you would use single quotes.
In Whatever Your Name Is, Netafura Yokohama approached to Gagohamo Mortimo with "angry eyes, and said, 'kiss my butt'."
Angry eyes, and said, "kiss my butt." is the direct quote, but since I already used double quotes to cite this quote, then the double quotes that surround kiss my butt becomes single quotes. There might be other uses, but that's the only one I know of.
