Hi,

Is  it  possible  to  ask  "What  is  he"  about  a  person's  job?

Yes. But in my experience, people usually ask 'What does he do?'

Hi,

For example What does he do for a living?

Yes. Although in contexts where you are clearly getting to know someone, the 'for a living' is often omitted.
