It has no particular religious significance. It's just the way you play the game.

As I recall, each player has a differnt pattern of numbers in the "boxes" on his card.
Someone calls off random numbers, one at a time.
Each player marks them as they happen to fall on his card.
The winner is the first to check off a particular pattern of boxes on his card.

It's just like saying "playing bingo", because (at least in some versions of the game) the numbers on the card, which appear in boxes on the card, are checked if they match the numbers called.
