Omar Ahmed1. Humanity has only one thing left, "hope."

The quotation marks have no place there. They are wrong.

Omar Ahmed2. Humanity has only one thing left: hope.

The colon is not called for. It is defensible but essentially wrong. The colon is best reserved for times when nothing else will do.

What is the correct punctuation to this sentence? I am really perplexed.

Humanity has only one thing left—hope.

The dash is better, but the comma suffices. The dash does what you were trying to do with the colon.
