'the' is the correct answer.

Here 'a' will suggest more than one road. Am I right?

Why 'a' is not the correct answer? The words 'one' and 'a' have very similar meanings and functions here. Don't use both.
'One' here is more colloquial and emphatic than 'a'.

3) Q: Is this __ road to San Francisco?

'the' is the correct answer. 'the'. You need an article.
