1) A man leans against a building, smoking a cigarette.

2) A man stands leaning against a building, smoking a cigarette.

If the man is standing, is sentence 1 enough or should I choose sentence 2?

1 is enough. I don't recommend 2.

Standing and leaning are two different actions. Standing means there is no support. For leaning, the wall is used as a support.
